Windows 7, Re Installed Avast 6.0.1, message indicating "invalid file signature", can not up date via the manual virus definition update method.
Then a RED pop up indicates that updates will not take.
Please advise.

Uninstall Avast completely. Run the Avast uninstaller. Download and install Avast Free 6.0.1044.

Did as you suggested in order, uninstall the 6.0.1, and reboot and then install 6.0.1044 as Administrator.
and the same issue arises.
This time the definitions are March 23, (with 6.0.1, the definitions were dated March 24, 0731am) and I can not update the virus definitions without the "invalid file signature" showing up.
Curious.
Please advise.

Are you sure your clock is on the current time? Did you run the Avast uninstaller?

Yes, I followed the link you indicated and downloaded the uninstaller, uninstalled Avast and then reboot, then
I installed Avast 601044.
The clock on my computer is linked to Windows 7 at the moment. Clock and date are showing current EDT of approximately 1042pm.
IN Avast the time of the most recent virus definitions is showing as: 3/23/11, 1:14:18PM.
Please advise.

Latest virus definitions also related to the march 23 date are 110323-1
and the message shows that my virus definitions are out of date and the latest definitions are 110329-1

I must go offline until Wednesday morning NYC time, about 9 hours from now. How concerned ought I be about my system NOT being protected?
The task bar icon shows Avast and says my system is secured. Which is it? Is my system secure or is it not secure?

Uninstalled Avast 6.01044, from SAFE MODE and reboot, reinstalled same in SAFE mode.
6.0.1044 Seems to be working now as the latest virus definitions showing are: 110330-0, and the date of update for virus definitions is March 30 (today's date) and the time showing for updating the definitions seems current.
Hope this helps, though I am still unsure what happened.
Avast has been my AV for many years, about 5 or 6 now and I am appreciative that it exists in the freeware version.

The newest version of Avast 6.0.1044, and the problem of not allowing virus updates persists.
Though the virus definitions were accepted upon installation, the definitions will not allow for update.
Same message: Invalid File Signature.
The most recent definitions are as posted above 110330.0, March 30, 2011.
Today is march 31, 2011. I get a RED pop up indicating the virus definitions are out of date. Manual update fails all the time: Invalid File Signature.
Please help!!!! Please advise. What is best to do,
and Yes, I did uninstall via the uninstall link above and reboot and reinstalled Avast 6.0.1044, several times, all from the SAFE MODE, administrator level.

What are the settings in "update"?
Select "no proxy" and "always connected".

You don't have to do it in Safe Mode if the normal way works...
I suggest an installation from scratch:
1. Download the latest version of avast! Uninstall Utility (http://www.avast.com/eng/avast-uninstall-utility.html) and save it.
2. Download the latest avast! (http://www.avast.com/eng/programs.html) version and save it.
3. Uninstall avast from Control Panel (if possible). If, for any reason, you can't run it, try booting in Safe Mode (http://www.pchell.com/support/safemode.shtml) and doing it from there. Anyway, boot after that.
4. Run the avast! Uninstall Utility saved on 1. If, for any reason, you can't run it, try booting in Safe Mode (http://www.pchell.com/support/safemode.shtml) and doing it from there. Anyway, boot after you've run it.
5. Install avast! using the setup saved on 2. Boot.
6. Register your free copy or add the license key for Pro. Or even upgrade (http://www.avast.com/resend-license-paid.php) your key from old versions.
7. Check and post the results. If, for any reason, you did not solve, try doing the step 3 in Safe Mode anyway.
Any other security program that could interfere?

My freeware version is registered and all that.
However, even with the adjustments in Settings, to NO Proxy, and Always Connected, the attempted manual update failed once again: invalid file signature. 13s this time to failure.
I have tried several times to install Avast 6.0.1044, from Normal Mode to Safe Mode. And yes, I have FIRST uninstalled only after using the uninstaller and rebooting.
The number of times I have done this is now nearing ten, I repeat ten (10). This seems a bit crazy.
Have been an avid Avast believer and user for many years.
In the past (you will see from my much earlier posts, a few years ago) there have been a glitch here and there, and this latest glitch is quite frustrating.
Is there a diagnostic I can run and post to allow for inspection by the powers that be, knowers of all knowledge Avast?
Am sure the issue is the result of a simple click here or check there, but which is it?
I can not find myself uninstalling and reinstalling Avast daily.

Fully understand your frustration.
However, this is not a common or widely spread behaviour, so I'ld suggest to have your PC scanned by Malwarebytes Antimalware, just to make sure it is not any malware that prevents the updates.
MBAM Quick Scan should be sufficient.

Something positive seems to have occurred.
Ran the freeware version of Malwarebytes Antimalware, and 3/4 of the way through, my system froze, (it found some malware related to what seemed to be good product: Java,
and after a reboot, the Avast updated correctly.
After that same reboot, ran Lavasoft Adaware also, and it found a few malware cookies.
Ran Malwarebytes again and system seems clean, for now.

After latest reboot due to Windows 7 IE 9.0 update, when I manually checked Avast update, the message indicated my engine and virus definitions are up to date.
Just ran manual update once again and all seems fine: and the No proxy setting and always on setting are in place.

It was only a stray registry key. Nothing was actually running or installed.

I have no log for that first pass with Malwarebytes as my computer crashed during the Malwarebytes process.
But I noticed that Java showed up several times with red, supposedly dangerous issues.
Those issues are gone anyway, now.
Nonetheless, the system is working well at the moment and if it ain't broke, I am not going to fix it.
